Characterization and elimination of a factor in serum that interferes with turbidimetry and nephelometry of lipase.

چکیده
We studied a serum component ("pseudolipase") that interferes with both turbidimetric and nephelometric lipase measurements. Pseudolipase activity in serum was found in the presence of rheumatoid factor and in other disorders of active immune processes. The incidence of this interferent in sera seems to be greater than reported previously. We describe a method for eliminating its effect without affecting pancreatic lipase activity, by precipitating some of the proteins, including IgM, with polyethylene glycol.

To assess interferences of carboxylester hydrolase (EC 3.1.1.1), which can cleave some pancreatic lipase substrates (4), we added 12 200 U of porcine liver carboxylesterase cat. no. E 3128) per liter to the reaction mixture and found no increase in lipase activity. negative values with the triolein assay, were also determined with the UV kinetic assay. In 28 samples, a value <5 U/L was observed...

Lipases are diversified enzymes in their properties and substrate specificity, which make them attractive tools for various industrial applications. In this study, an alkaline thermostable lipase producing bacteria were isolated from soil of different regions of Isfahan province (Iran) and its lipase was purified by ammonium sulfate precipitation and ion exchange chromatography. To select a the...
متن کاملMolecular Cloning and Characterization of a Lipase from an Indigenous Bacillus pumilus
Cloning and sequencing of a lipase gene from an indigenous Bacillus pumilus, strain F3, revealed an open-reading frame of 648 nucleotides predicted to encode a protein of 215 residues. Sequence analysis showed that F3 lipase contained a signal peptide composed of 34 amino acids with an H domain of 18 residues. A tat-like motif was found in the signal peptide similar to some other Bacillus pumil...

متن کاملIsolation, Optimization, and Molecular Characterization of a Lipase Producing Bacterium from Oil Contaminated Soils
Lipases have many applications in biotechnology, thanks to their ability of acylglycerides hydrolysis. They alsp possess the unique feature of acting at the lipid-water interface, which distinguishes them from esterases. Commercially useful lipases are produced by microorganisms with the extracellular lipase being produced by many bacteria including Pseudomonas. The greatest production of lipas...
